Removing Counters by Damage
Question,
If my mycoloth has 6 +1/+1 counters, and i take 5 damage, do i lose 5 counters? or do i lose 1 counters (mycoloth is a 4/4)..
general question is,
do counters regenerate like toughness does at the end of turn.
if they do not regen, what gets damaged first counters or toughness

it seems you do not understand damage
when a creature is dealt damage, lets say the creaure is a 6/6 and is dealt 4 damage, the creature is still a 6/6, it just took 4 damage that turn and the damage lasts until the turn is over (damage is not represented by counters!!).
So your mycoloth never looses any counters if it takes damage.
If a 6/6 takes a lighting bolt (3 damage) and the blocks a 4/4 the 4/4 dies because it gets 6 damage from the 6/6 and the 6/6 dies because it took 3 + 4 damage in the same turn.
What you are referring to sounds like "wither". Creatures with the wither mechanic deal their damage in the form of -1/-1 counters. Now counters unlike damage are permanent and -1/-1 counters and +1/+1 counters cancel eachother out.
so if a creature with wither deals 4 damage to mycoloth your mycoloth with 4 +1/+1 counters on it looses 4 counters permanently.

thanks for the clarification,
i had asked another friend of mine in the meantime and he had settled the questions by explaining that like unholy strength where it enchants with a +2/+1, a +1/+1 counter is like an enchantment and dealing damage doesn't remove enchants (i do understand though that counters are not enchantments)
