It kills one defender for each point of power it has.
EX: A 1/1 Wolf with Deathtouch blocked by 3 creatures will kill just 1, while a 2/2 Deathtouch Wolf will kill 2, and one with 3 or higher power will kill them all.

You are correct! Be it combat damage, or damage from it's own effects, Deathtouch will be satisfied and come into play.

Exceptions are as follows.
-Anything that has protection from creatures/red/elementals/hounds/everything because the damage would be prevented.
-Anything that specifically prevents all damage to it.Phantom Flock
-Anything Indestructible.(Deathtouch is a destroy effect)
You can use cards like Flaring Pain to get around protection effects, but not indestructible.
